1.0 PURPOSE

The purpose of the standard operating procedure is to provide guidelines for calibration of Inductively Coupled Plasma Mass Spectrometer.

2.0 SCOPE

The scope of this standard operating procedure includes calibration of Inductively Coupled Plasma Mass Spectrometer.

3.0 RESPONSIBILITY

3.1 Application Scientist

3.2 Technical-Manager

3.3 Q.A. Manager

4.0 REQUIREMENTS

4.1 Argon Gas Cylinders

4.2 Elan 9000 Hardware Guide

4.3 Elan Inductively Coupled Plasma Mass Spectroscopy safety manual

4.4 Tuning Solution

5.0 PROCEDURE

5.1 Precaution while start-up the Inductivel Coupled Plasma Mass Spectrometer

5.1.1 Argon gas cylinder should not be used below 100 psi in term of gas quantity, Release gas pressure should be between 60-80 psi.

5.2 Switch on the blower & recirculater.

5.3 Before the ignition of the plasma, vacuum pressure should be 10-6

5.4 Open Software.

5.4.1 Open “Instrument “———Start Plasma

5.4.2 Leave the plasma for 30 minutes or more (minimum 25 minutes).

5.4.3 Open —-“ Workspace” ———-“Daily Performance .wrk”

5.4.4 Place the tuning solution.

5.4.5 Analyse the above solution as “Sample “.

5.4.6 Do Daily Performance Check

    • Mg intensity > 40,000 cps/10 ppb
    • In Intensity > 250 , 000 cps/10 ppb
    • U intensity > 200,000 cps/10 ppb
    • CeO/Ce ;Ba++/Ba < 3 %
    • Background 220 <2 cps

5.4.7 If above test is not OK then optimize. Optimisation is required only when the value are highly disturbed .Do optimization while referring to the manual. following parameters must be optimized.

5.4.7.1 Nebuliser Gas Flow (NEB)

5.4.7.2 Auxiliary Gas Flow

5.4.7.3 Plasma Gas Flow

5.4.7.4 Lens Voltage

5.4.7.5 Inductively Coupled Plasma Radio-frequency (Rf) power

5.4.7.6 Analog Stage voltage

5.4.7.7 Pulse Stage voltage

5.4.7.8 Quadropole Rod Offset (QRD)

5.4.7.9 Cell Path Voltage (CPV)

5.5 Click on get Analyte list Icon & select Analyte from Analyte Icon i.e.Mg

5.6 Select optimization criteria & select maximum intensity.

5.7 Click on optimize icon.

5.8 Repeat the step 5.4.8 , 5.4.9 & 5.4.10 for above mentioned parameters individually i.e. Nebuliser Gas Flow, Auxiliary Gas Flow, and Plasma Gas flow
